چکیده

Genetic and environmental variation in carbon isotope discrimination (A), photosynthetic gas exchange, growth and activities of phosphoenolpyruvate carboxylase (PEPC) and ribulose-1,5-bisphosphate carboxylase (Rubisco) were studied in four sugarcane clones grown under three different irrigation regimes for 65 d in a greenhouse. A of the uppermost fully expanded leaf increased with decreasing irrigation frequency and exhibited genetic variation at all irrigation frequencies. Concurrent measurements of leaf gas exchange characteristics suggested that variation in \ was attributable to variation in bundle sheath leakiness to CO2 (<D) because the ratio of intercellular to ambient partial pressure of CO2 remained relatively constant except at the lowest level of water availability. Carboxylase assays performed on two genotypes revealed significant genetic variation in Rubisco activity, but no genetic variation in PEPC activity. The Rubisco:PEPC activity ratio, an index of C3:C4 cycle activity, therefore varied genetically. Rubisco: PEPC activity declined significantly with decreasing irrigation frequency. Both A and 0 were negatively correlated with genetic and environmental variation in Rubisco: PEPC activity. Taken together, these results suggest that variation in A in sugarcane is governed largely by relative leakage of CO2 from the bundle sheath mediated by changes in Rubisco:PEPC activity. C0 2 assimilation (4) and shoot growth rate [SCR) were negatively correlated with A and <P, but positively correlated with Rubisco: PEPC activity. Thus, A can serve as an integrated measure of underlying variation in photosynthetic biochemistry associated with variation in performance at the whole plant level in sugarcane and probably other C4 species.

Photosynthetic flexibility in maize exposed to salinity and shade

C4 photosynthesis involves a close collaboration of the C3 and C4 metabolic cycles across the mesophyll and bundle-sheath cells. This study investigated the coordination of C4 photosynthesis in maize plants subjected to two salinity (50 and 100mM NaCl) treatments and one shade (20% of full sunlight) treatment. Photosynthetic efficiency was probed by combining leaf gas-exchange measurements with...

Elevated CO(2) induces biochemical and ultrastructural changes in leaves of the C(4) cereal sorghum.

We analyzed the impact of growth at either 350 (ambient) or 700 (elevated) microL L(-1) CO(2) on key elements of the C(4) pathway (photosynthesis, carbon isotope discrimination, and leaf anatomy) using the C(4) cereal sorghum (Sorghum bicolor L. Moench.). Gas-exchange analysis of the CO(2) response of photosynthesis indicated that both carboxylation efficiency and the CO(2) saturated rate of ph...

Antisense Reduction of NADP-Malic Enzyme in Flaveria bidentis Reduces Flow of CO2 through the C4 Cycle [W][OA]

An antisense construct targeting the C4 isoform of NADP-malic enzyme (ME), the primary enzyme decarboxylating malate in bundle sheath cells to supply CO2 to Rubisco, was used to transform the dicot Flaveria bidentis. Transgenic plants (a-NADP-ME) exhibited a 34% to 75% reduction in NADP-ME activity relative to the wild type with no visible growth phenotype. We characterized the effect of reduci...

Growth of the C4 dicot Flaveria bidentis: photosynthetic acclimation to low light through shifts in leaf anatomy and biochemistry

In C(4) plants, acclimation to growth at low irradiance by means of anatomical and biochemical changes to leaf tissue is considered to be limited by the need for a close interaction and coordination between bundle sheath and mesophyll cells. Here differences in relative growth rate (RGR), gas exchange, carbon isotope discrimination, photosynthetic enzyme activity, and leaf anatomy in the C(4) d...
